Uses for L-Glutamine

The synthetic method of L-glutamine
L-glutamine widely exists in nature, for example, it is contained in pumpkin and sunflower seedlings in a free state, and its N-ethyl compound (theanine, theanine) is contained in tea leaves. While glutamine can be extracted from natural products, it is produced in large quantities by fermentation and synthesis.
1. Synthesis method
The is obtained from L-glutamic acid-5-methyl ester ([1499-55-4]) through condensation, addition, salt formation and hydrolysis. Glutamic acid is esterified with methanol in the presence of concentrated sulfuric acid, and the resulting esterified solution is added dropwise into a mixture of methanol and carbon disulfide, and ammonia is passed under cooling while adding dropwise. After the esterification solution was added dropwise, continue to pass through ammonia, then add triethylamine, and place it in a closed place at 30°C for 40h. After being concentrated under reduced pressure to catch ammonia, the concentrated solution of γ-methyl ester-L-glutamic acid-N-diammonium carbamate was obtained. It was heated to 40-45°C and acetic acid was added. After stirring for 30 min, carbon disulfide was removed under reduced pressure, and a large amount of crystals were precipitated at this time. Then add an equal volume of methanol, place at 0°C for 12h, filter to obtain crude glutamine. Decolorized by activated carbon and recrystallized to obtain the finished product.
2. Fermentation method
Glucose, acetic acid, and ethanol are used as the carbon source of the medium, and Brevibacterium flavum is used for fermentation. The yield based on glucose is 39g/L, and the yield is 39%.
